Hard to tell if it's a step up in quality, you'll need to do some a/b testing if you like it or not.
But you'd connect the 6i6 Line Out to the Kemper TRS return with a 1/4" TRS (balanced) cable, and turn down the input sensitivity from the KPA.
A great preamp can help to get a better profile - but in most cases is the influence of the mic and mic position much bigger.
I would also to profile the amp with the inbuilt mic pre and then use the external one without changing anything with your mic and mic position - then compare - use the preamp you prefer.
In case you use a phantom powered mic in the future - then you need a good mic pre with phantom power (the KPA does not deliver phantom power).
I also found that I prefer the sound of the R121 much more via external preamps then via the KPA inbuilt one.

I thought it was just a balanced line input? The Return Level isn't an analog gain control, is it?
