Can anyone with experience post up on the tonal effect of profiling with a cab with new vs broken in speakers? As broken in speakers sound better, I'm curious if it effects the profiled tone or not.

Which difference do you hear in new vs. broken-in speakers?
For studio-profiles I'd suggest to wait 'til the Speakers are broken in. It's a huge difference. Broken-in-Speakers are much louder than brand new ones and also the frequency-range is much more expanded. A good solution for breakin' in new Speakers - when there's no time to do this by playin' throught them, is to use an ac-transformer f.e. Märklin-model and let it run 20 - 30 hours.
Though this causes really good results for the Speaker-Sound your matrimony may suffer, when you do this at home.
If you like the sound of non broken in speakers this is them time to profile.
If you do not, break them in first.
For me some speakers do not need to be broken in, others are horrid until they are.

In all seriousness, I agree you should break in your speakers first. That's when they sound their best.
