Noob Question: Can the Kemper profile a plugin EQ

You need two Kempers to do this. One Kemper will be set to the sound of the desired profile, which will run into Isotope. The other will be used to profile this setup.

You do not need two KPAs to do that. You you need a source amp, either DI or miked, and use ozone on that amp signal and take a profile of the target signal. Using a tube amp as source signal that has a gain structure and dynamics similar to your target amp is highly recommended. Some people use modellers or software amps as source signals. Imo using a real tube amp has some major benefits. You keep all the dynamics and behaviour of the original amp and use ozone only to eq the signal.

In my defence, I'll point out that he specifically mentioned the Lasse 5150 amp, which is not available to me at least outside the Kemper. Naturally, you can have an EQ in the signal chain when profiling an amp
