For those of you who are experienced with this, is there a big impact in profiling results with an amp with old worn-out tubes versus new tubes? Is the difference just as noticeable as playing a freshly retubed amp?

From what I've heard, yes!
After all, the Kemper will capture what's there. If the tubes are clapped-out, that's what you'll end up with for posterity. Not ideal of course, mate.

If the amp sounds/feels different with worn out tubes than new tubes, the profile will, too.
The same is true of over/under biased poweramp tubes, preamp tube substitutions, etc.

Absolutely. Tubes change over time, some of my favorite old amps lost their tone and dynamics that I loved after the tubes wore a bit. Finding NOS or decent replacements didn't always work out.
